    Deep Engagement with Student Learning: Librarians as Instructors-of-Record for Writing-Intensive Undergraduate Courses

    How can librarians gain authentic knowledge about how students apply the skills and concepts we teach? In order to address this question, Susan Barnes Whyte and Jean Caspers share the knowledge they have gained by teaching two writing intensive courses: Information Gathering, a required 4-credit course for Mass Communication majors, and Information Ethics: the Individual as Creator and Consumer, a required 4-credit first year inquiry seminar course. Caspers and Whyte both continue to teach multiple information literacy (IL) sessions for other professors’ courses. Their for-credit teaching experiences have helped them understand the difficulty other teaching faculty have finding time to integrate IL into their courses, but have also underlined the necessity for students to integrate IL concepts into their research and writing practice. Successful negotiation for more time and more in-depth assignments in all the classes they touch has been an important and positive outcome for their work

    Splittings of generalized Baumslag-Solitar groups

    We study the structure of generalized Baumslag-Solitar groups from the point of view of their (usually non-unique) splittings as fundamental groups of graphs of infinite cyclic groups. We find and characterize certain decompositions of smallest complexity (`fully reduced' decompositions) and give a simplified proof of the existence of deformations. We also prove a finiteness theorem and solve the isomorphism problem for generalized Baumslag-Solitar groups with no non-trivial integral moduli.Comment: 20 pages; hyperlinked latex.     Efficient, Blind, Spatially-Variant Deblurring for Shaken Images

    International audienceIn this chapter we discuss modeling and removing spatially-variant blur from photographs. We describe a compact global parameterization of camera shake blur, based on the 3D rotation of the camera during the exposure. Our model uses three-parameter homographies to connect camera motion to image motion and, by assigning weights to a set of these homographies, can be seen as a generalization of the standard, spatially-invariant convolutional model of image blur. As such we show how existing algorithms, designed for spatially-invariant deblurring, can be "upgraded" in a straightforward manner to handle spatially-variant blur instead. We demonstrate this with algorithms working on real images, showing results for blind estimation of blur parameters from single images, followed by non-blind image restoration using these parameters. Finally, we introduce an efficient approximation to the global model, which significantly reduces the computational cost of modeling the spatially-variant blur. By approximating the blur as locally-uniform, we can take advantage of fast Fourier-domain convolution and deconvolution, reducing the time required for blind deblurring by an order of magnitude

    Reset-free Trial-and-Error Learning for Robot Damage Recovery

    The high probability of hardware failures prevents many advanced robots (e.g., legged robots) from being confidently deployed in real-world situations (e.g., post-disaster rescue). Instead of attempting to diagnose the failures, robots could adapt by trial-and-error in order to be able to complete their tasks. In this situation, damage recovery can be seen as a Reinforcement Learning (RL) problem. However, the best RL algorithms for robotics require the robot and the environment to be reset to an initial state after each episode, that is, the robot is not learning autonomously. In addition, most of the RL methods for robotics do not scale well with complex robots (e.g., walking robots) and either cannot be used at all or take too long to converge to a solution (e.g., hours of learning). In this paper, we introduce a novel learning algorithm called "Reset-free Trial-and-Error" (RTE) that (1) breaks the complexity by pre-generating hundreds of possible behaviors with a dynamics simulator of the intact robot, and (2) allows complex robots to quickly recover from damage while completing their tasks and taking the environment into account. We evaluate our algorithm on a simulated wheeled robot, a simulated six-legged robot, and a real six-legged walking robot that are damaged in several ways (e.g., a missing leg, a shortened leg, faulty motor, etc.) and whose objective is to reach a sequence of targets in an arena.     Structure of the Mg-Chelatase Cofactor GUN4 Reveals a Novel Hand-Shaped Fold for Porphyrin Binding

    In plants, the accumulation of the chlorophyll precursor Mg-protoporphyrin IX (Mg-Proto) in the plastid regulates the expression of a number of nuclear genes with functions related to photosynthesis. Analysis of the plastid-to-nucleus signaling activity of Mg-Proto in Arabidopsis thaliana led to the discovery of GUN4, a novel porphyrin-binding protein that also dramatically enhances the activity of Mg-chelatase, the enzyme that synthesizes Mg-Proto. GUN4 may also play a role in both photoprotection and the cellular shuttling of tetrapyrroles. Here we report a 1.78-Å resolution crystal structure of Synechocystis GUN4, in which the porphyrin-binding domain adopts a unique three dimensional fold with a “cupped hand” shape. Biophysical and biochemical analyses revealed the specific site of interaction between GUN4 and Mg-Proto and the energetic determinants for the GUN4 • Mg-Proto interaction. Our data support a novel protective function for GUN4 in tetrapyrrole trafficking. The combined structural and energetic analyses presented herein form the physical-chemical basis for understanding GUN4 biological activity, including its role in the stimulation of Mg-chelatase activity, as well as in Mg-Proto retrograde signaling

    Mild forms of hypophosphatasia mostly result from dominant negative effect of severe alleles or from compound heterozygosity for severe and moderate alleles

    <p>Abstract</p> <p>Background</p> <p>Mild hypophosphatasia (HPP) phenotype may result from <it>ALPL </it>gene mutations exhibiting residual alkaline phosphatase activity or from severe heterozygous mutations exhibiting a dominant negative effect. In order to determine the cause of our failure to detect a second mutation by sequencing in patients with mild HPP and carrying on a single heterozygous mutation, we tested the possible dominant effect of 35 mutations carried by these patients.</p> <p>Methods</p> <p>We tested the mutations by site-directed mutagenesis. We also genotyped 8 exonic and intronic <it>ALPL </it>gene polymorphisms in the patients and in a control group in order to detect the possible existence of a recurrent intronic mild mutation.</p> <p>Results</p> <p>We found that most of the tested mutations exhibit a dominant negative effect that may account for the mild HPP phenotype, and that for at least some of the patients, a second mutation in linkage disequilibrium with a particular haplotype could not be ruled out.</p> <p>Conclusion</p> <p>Mild HPP results in part from compound heterozygosity for severe and moderate mutations, but also in a large part from heterozygous mutations with a dominant negative effect.</p

    Efficient TALEN-mediated gene targeting of chicken primordial germ cells

    In this work we use TALE nucleases (TALENs) to target a reporter construct to the DDX4 (vasa) locus in chicken primordial germ cells (PGCs). Vasa is a key germ cell determinant in many animal species and is posited to control avian germ cell formation. We show that TALENs mediate homology-directed repair of the DDX4 locus on the Z sex chromosome at high (8.1%) efficiencies. Large genetic deletions of 30 kb encompassing the entire DDX4 locus were also created using a single TALEN pair. The targeted PGCs were germline competent and were used to produce DDX4 null offspring. In DDX4 knockout chickens, PGCs are initially formed but are lost during meiosis in the developing ovary, leading to adult female sterility. TALEN-mediated gene targeting in avian PGCs is therefore an efficient process

    Endovascular coils as lung tumour markers in real-time tumour tracking stereotactic radiotherapy: preliminary results

    To evaluate the use of endovascular coils as markers for respiratory motion correction during high-dose stereotactic radiotherapy with the CyberKnife, an image-guided linear accelerator mounted on a robotic arm. Endovascular platinum embolisation coils were used to mark intrapulmonary lesions. The coils were placed in subsegmental pulmonary artery branches in close proximity to the target tumour. This procedure was attempted in 25 patients who were considered unsuitable candidates for standard transthoracic percutaneous insertion. Vascular coils (n = 87) were succesfully inserted in 23 of 25 patients. Only minor complications were observed: haemoptysis during the procedure (one patient), development of pleural pain and fever on the day of procedure (one patient), and development of small infiltrative changes distal to the vascular coil (five patients). Fifty-seven coils (66% of total inserted number) could be used as tumour markers for delivery of biologically highly effective radiation doses with automated tracking during CyberKnife radiotherapy. Endovascular markers are safe and allow high-dose radiotherapy of lung tumours with CyberKnife, also in patients who are unsuitable candidates for standard transthoracic percutaneous marker insertion
